Production of in vitro embryos (PIV), with oocytes obtained by opu of heifers stimulated with alternative protocols

Project: Research

Project Details

Description

The experiment will be carried out in the Irquis experimental farm of the Faculty of Agricultural Sciences of the University of Cuenca, seeks to establish alternative protocols for ovarian stimulation based on FSH-LH, to obtain bovine oocytes through the OPU technique, which allow improving the in vitro (PIV) embryo production process. 9 manstizas holstein genotype heifes will be used, under grazing handling system, 3 experiments will be applied in these animals, the first seeks to determine the emergency time of the follicular wave after the application of three treatments based GNRH (0.2mg im), stradiol benzoate (2mg im) and follicular ablation (follicles> 5mm), for the assessment they will be determined every 24h Folicular dynamics through ultrasound and blood FSH levels. The second study will allow evaluating the response to ovarian stimulation with FSH-LH, prior to the Opu session in terms of the follicular response, quantity and quality of recovered cocs, for which the number of follicles present before the OPU, the quantity and quality of recovered coc´s will be determined. Finally, it is intended to standardize the in vitro (PIV) embryo production process through the use of oocytes obtained by OPU, the process will be developed in microgotes with the standardized media and times in the Biotechnology Laboratory for the PIV with oocytes from slaughterhouse. For the statistical analysis, the SAS program will be applied, socks and frequencies will be determined and for the comparison of means the Tukey-Kramer test will be applied.
