Description
The prevalence of children's asthma is very high around the world. Its complexity responds to several associated environmental and genetic factors. Although many cases of asthma are explained by genetic factors, epigenetic changes constitute plausible necanisms for their development. Studies of epignetic variations associated with asthma are scarce in developing countries. The objective is to indicate the genes associated with asthma when comparing the methylation patterns associated with these genes between asthmatic children and healthy children. In addition, associate the degree of methylation with severity in its symptomology. A study of cases and controls will be carried out for the identification of epigenetic variations associated with asthma. 80 cases will be identified in the Pneumology Service and 80 controls in the Pediatrics Services of Monte Sinaí Hospital. In the DNA from saliva samples, methylation patterns will be identified using the Infinium Humanmethylation450 Beadchip Array (Illumina, San Diego, CA, CA, USA) commercial panel in the Genetics Laboratory of the University of Belgian Antwerp by Dr. Wilm Vanden Berghe PhD. The analyzes will be carried out according to the methylation protocol of the infinium HD test. Guide (Part · 15019519, Illuminina). At the end of this research, it is expected to find regions with methylation differences in the genes associated with asthma that allow to establish differences between healthy individuals and with the disease. In addition, techniques could be established to predict and diagnose the disease in their early stages and avoid its progression to severe stages.
